The use of dextran sulphate as a blood anticoagulant in biological research.

چکیده
Several well-known anticoagulants are in use for blood sample tubes, each having some special application, but there is little doubt that the naturally occurring anticoagulant, heparin, would find wider application were it not so difficult to prepare and consequently expensive. Chemically prepared sulphuric esters of polysaccharides are known to have anticoagulant action. One of these, dextran sulphate, has recently been prepared (Ricketts, 1952) in a form suitable for therapeutic use (Walton, 1951). This material, which is readily prepared and therefore cheaper than heparin, may also find application in biological laboratory work. Some experiments have been conducted to ascertain whether dextran sulphate will give results comparable with the usual anticoagulants in determinations commonly performed in the clinical pathology laboratory. While not comprehensive, these experiments indicate the general suitability of the material for this purpose.

Dextran sulphate: the relation of molecular features to biological properties.
In a previous paper (Ricketts, 1952a) a series of dextran sulphate preparations varying widely in molecular weight and sulphur content was described. The biological properties of these compounds were examined by Walton (1951, 1952), who found that molecular weight was the principal feature determining toxicity, the smaller molecules being the least toxic. At molecular weights (MN) of the order ...
متن کاملProtective Effects of Pioglitazone against Dextran Sodium Sulphate-Induced Colitis in Rats.
The aim of the present study is to investigate the preventive effect of pioglitazone on colitis induced with dextran sulfate sodium (DSS) in rats. Twenty-four male Sprague-Dawley rats weighing 180-200 g were randomized into four groups. Rats of the 1st group received only saline and served as normal group. Colitis was induced in the remaining 3 groups by 1.5% DSS administered in drinking water ...
متن کاملPartial Purification and Characterization of Anticoagulant Factor from the Snake (Echis carinatus) Venom
Objective(s): Snake venoms contain complex mixture of proteins with biological activities. Some of these proteins affect blood coagulation and platelet function in different ways. Snake venom toxin may serve as a starting material for drug design to combat several pathophysiological problems such as cardiovascular disorders. In the present study, purification of anticoagulation facto...
متن کاملThe estimation and recovery of dextran sulphates in biological fluids.
The dextran sulphates resemble heparin in being active inhibitors of several different forms of biological activity, e.g., blood coagulation, serum complement activity, and certain enzyme systems (Walton, 1954). In each case the activity of the dextran sulphate appears to be dependent upon direct interaction with one or more protein components of the biological system under investigation. In fa...

The case of a 34 year old woman with systemic lupus erythematosus with a history of three previous recurrent abortions and lupus anticoagulant and anticardiolipin antibodies is reported. Immunoadsorbent plasmapheresis with a dextran sulphate column was used to remove lupus anticoagulant, anticardiolipin antibodies, and antibodies to DNA during her fourth pregnancy in combination with low doses ...
